Anti-Inflammatory and Neuroprotective Effects of Opuntia ficus-indica var. saboten Alone or Combined with Vitamin C

Abstract
Opuntia ficus-indica var. saboten has long been known as a medicinal plant on Jeju Island in Korea for treatment of diabetes, asthma, burns, and dyspepsia. Although Opuntia ficus-indica var. saboten has been reported to have analgesic and anti-inflammatory effects in several studies, its mode of action has not been clearly described. In this study, we investigated the neuroprotective effects of ethanolic extract of Opuntia ficus-indica var. saboten (EOFS) on neuronal inflammation in vitro. We verified the nitric oxide levels and the decreased levels of inducible nitric oxide synthase and cyclooxygenase-2 expression to assess its effects on inflammation in macrophage cells. We also performed an acetylcholinesterase (AChE) activity assay to validate the AChE inhibitory effects of EOFS. In addition, a cellular viability assay was performed to confirm the protective effects of EOFS against ¥â-amyloid-induced neurotoxicity. We found increased levels of cAMP response element-binding protein and brain-derived neurotrophic factor expression upon evaluation of the regulation of synaptic plasticity by EOFS in SH-SY5Y neuroblastoma cells. Furthermore, we verified the synergistic effects of combined treatment with EOFS and vitamin C. Consequently, these results suggest that EOFS could be used as a source of functional materials to improve the cognitive function generated by anti-inflammatory response, AChE inhibition, cellular protection, and regulation of synaptic plasticity.
